چکیده

To elucidate the role of p53/p16/RB1 pathways in the tumorigenesis of primary central nervous system lymphomas (PCNSLs), we have analyzed p14, p16, RB1, p21, and p27 status in a series of their 18 sporadic cases of diffuse large B-cell lymphoma, using methylationspecific PCR, differential PCR, and immunohistochemistry. Homozygous deletion or methylation of p14 was detected in 10 (56%) PCNSLs, and they were almost entirely deletions (except 1 case). A total of 11 (61%) PCNSLs demonstrated homozygous deletion (6 cases) or methylation (5 cases) of p16. Six tumors showed both p14 and p16 homozygous deletions. Hypermethylation of the RB1 and the p27 promoter region was detected in 2 (11%) cases, whereas p21 methylation was not detected in any. Immunohistochemistry revealed loss of p14 and p16 expression in 10 (56%) samples, correlating with the gene status. Four cases showed independent negative immunoreactivity for pRB and p27, and nearly one-half of cases (8 of 18; 44%) were characterized by lack of p21 expression. These results indicate that inactivation of p14 and p16 by either homozygous deletion or promoter hypermethylation represents an important molecular pathogenesis in PCNSLs. Hypermethylation of RB1, p21, and p27 appears to be of minor significance, these genes being independently methylated in PCNSLs.

Imaging, Diagnosis, Prognosis Prognostic Significance of p14, p15, and p16 Inactivation in Malignant Peripheral Nerve Sheath Tumors

Purpose: p14, p15, and p16 are tumor suppressor genes that are located closely at 9p21 and are often coinactivated by genetic or epigenetic alterations. Malignant peripheral nerve sheath tumor (MPNST) is a rare sarcoma with poor prognosis. However, the prognostic implications of inactivation of p14, p15, and p16 in MPNSTs have not been adequately investigated. Here we carried out a genetic, epi...

Frequent methylation silencing of p15(INK4b) (MTS2) and p16(INK4a) (MTS1) in B-cell and T-cell lymphomas.

The methylation status of p15(INK4b) (MTS2), p16(INK4a) (MTS1) and p14(ARF) (p16beta) was analyzed in 56 lymphomas by restriction-enzyme related polymerase chain reaction (PCR) (REP), methylation-specific PCR (MSP), and bisulfite genomic sequencing (BGS). Methylation of the p15 and p16 genes was detected, respectively, in 64% and 32% of the B-cell lymphomas, in 44% and 22% of the T-cell lymphom...

p14(ARF) nuclear overexpression in aggressive B-cell lymphomas is a sensor of malfunction of the common tumor suppressor pathways.

p14(ARF), the alternative product from the human INK4a/ARF locus, antagonizes Hdm2 and mediates p53 activation in response to oncogenic stimuli. An immunohistochemical study of p14(ARF) expression in 74 samples of aggressive B-cell lymphomas was performed, demonstrating an array of different abnormalities. A distinct nucleolar expression pattern was detected in nontumoral tissue and a subset of...

Advance and Briefs Mechanisms of Inactivation of p14, p15, and p16 Genes in Human Esophageal Squamous Cell Carcinoma

The 9p21 gene cluster, harboring growth suppressive genes p14, p15, and p16, is one of the major aberration hotspots in human cancers. It was shown that p14 and p16 play active roles in the p53 and Rb tumor suppressive pathways, respectively, and p15 is a mediator of the extracellular growth inhibition signals. To elucidate specific targets and aberrations affecting this subchromosomal region, ...

Promoter Methylation of RASSF1A Associates to Adult Secondary Glioblastomas and Pediatric Glioblastomas

While allelic losses and mutations of tumor suppressor genes implicated in the etiology of astrocytoma have been widely assessed, the role of epigenetics is still a matter of study. We analyzed the frequency of promoter hypermethylation by methylation-specific PCR (MSP) in five tumor suppressor genes (PTEN, MGMT, RASSF1A, p14(ARF), and p16(INK4A)), in astrocytoma samples and cell lines. RASSF1A...
